Figure 4.
A multi-panel image depicts data on CD36WT and CD36G12V molecules. Panel A shows boxplots of regression coefficients for the diffusion coefficient of CD36WT and CD36G12V molecules on local CA speckle displacement, categorized by low and high speckle density and free or confined states. The x-axis represents local speckle displacement, and the y-axis represents the regression coefficient. Panel B shows boxplots of regression coefficients for the diffusion radius of CD36WT and CD36G12V molecules on local CA speckle displacement, similarly categorized. Panel C shows boxplots of local CA speckle displacement magnitude at CD36WT and CD36G12V molecules in the center and periphery regions. The x-axis represents the region, and the y-axis represents the displacement magnitude. Panel D shows boxplots of the diffusion coefficient of confined CD36WT and CD36G12V tracklets in the center and periphery regions. The x-axis represents the region, and the y-axis represents the diffusion coefficient. Panel E shows boxplots of the diffusion radius of confined CD36WT and CD36G12V tracklets in the center and periphery regions. The x-axis represents the region, and the y-axis represents the diffusion radius. Each panel includes individual cell data points, the mean of the group of cells, and the standard deviation of the group of cells, with different colors representing CD36WT and CD36G12V molecules. Significant differences are indicated by asterisks and pictograms.

Confined CD36 G12V molecules exhibit less coupling to CA speckle displacement than confined CD36 WT molecules. (A and B) Regression coefficients of CD36WT and CD36G12V diffusion coefficient (A) and diffusion radius (B) on local CA speckle displacement, from MVRG analysis on speckle displacement and density (see Fig. S6 for corresponding regression coefficients on density). MVRG analysis was performed separately for the four indicated subgroups of SM tracklets based on local speckle density and diffusion type. In addition, next to the pictograms, +, ++, +++/−, --, and --- indicate significant differences (greater/smaller; P value ≤ 0.05, 0.01, and 0.001, respectively) between confined and free MVRG coefficient magnitudes at the same CA density level for each type of molecule. P value > 0.05 is not explicitly indicated to reduce clutter. (C) Local CA speckle displacement magnitude at CD36WT and CD36G12V molecules in the center and periphery regions. (D and E) Diffusion coefficient (D) and diffusion radius (E) of confined CD36WT and CD36G12V tracklets in the center and periphery regions. Circles, red lines, shaded bars, asterisks (or lack thereof), and pictograms (the latter in A and B) are as in Fig 2. See Table 5 for number of cells, number of experimental repeats, and number of SM tracklets and speckles per ROI used for analysis.
