MEFsv40 cells co-expressing Opto-CAAX-RFP/opto-β3 integrin-Venus, opto-ALK3-RFP/opto-β3 integrin-Venus, or opto-BMPRII-RFP/opto-β3 integrin-Venus were seeded onto glass before pSMAD 1/5 immunostaining. (A and C) Without sBMP2, opto-BMPR overexpression or blue light activation was not sufficient to induce the phosphorylation and translocation of SMAD 1/5 to the nucleus.
