Less than 10% of CD59 cluster rafts and Ab-CTXB-GM1 clusters became trapped in caveolae within 10 min after their induction.
(A and B) The fractions of CD59 cluster rafts (A) and Ab-CTXB-GM1 clusters (B) colocalized with the caveolin-1–GFP spots, as detected by simultaneous, two-color imaging at single-molecule sensitivity. The colocalized fractions increased with time but remained at <10% within 10 min after the cluster formation initiation. Based on these results, all of the experiments for observing the recruitment of intracellular molecules at CD59 cluster rafts and Ab-CTXB-GM1 clusters were performed within 10 min after cluster induction.
