Cells expressing the APVNTT-DsRed-Express2-FKBP^RD(C22V) cargo together with Sec7-HaloTag and Apl2-GFP were grown to mid-log phase, labeled with JF₆₄₆, treated with SLF for 5 min, and imaged by 4D confocal microscopy with an interval of 1 s between Z-stacks.
