A shift in the gating transition state indicates there is a local conformational change at 9′. Change in forward gating rate constant log(δf0) versus the change in gating equilibrium constant log(δE0) (REFER) for different mutations at the indicated side chain positions (9′, 12′, and 13′). The slope of the regression line (ϕ) indicates the position of the local conformational change relative to the global transition state (TS or committer) on a scale from 1 to 0. ϕ = 1, open-like conformation at the TS; ϕ = 0, closed-like conformation at the TS. Intermediate values of ϕ represent intermediate positions along the C↔O pathway. The average ϕ value for the 9′, 12′, 13′, and 16′ residues = 0.55.
