(A) Gating is shown on a representative spleen sample but is also applicable to gating in the BM, dura, LM, and brain. The same gating is used for blood samples with the exception that double-positive CD45 and CD45 i.v. fractions were used for subsequent analysis. (B) Representative gating of CFSEhigh and CFSElow CD3+ T cells is shown at both preclinical (day 1 after A/T) and clinical (day 7 after A/T) time points of EAE. (C) CFSEhigh (blue) and CFSElow (green) CD3+ T cells were quantified throughout EAE in the spleen, the brain, and brain borders as indicated. Results in C are data from three independent experiments. A total of 4–12 mice were used per time point, as indicated in graph legends. Error bars indicate mean ± SEM. Statistical analysis was not conducted on this data set.
