Proposed schematic events targeting LD catabolism by ethanol. p97/VCP is recruited to the LD surface by UBXD8, which extracts HSD17β13 from the LD and delivers HSD17β13 to the lysosomes via interaction with HSC70 and LAMP2A for chaperone-mediated autophagic degradation of HSD17β13. Alcohol consumption decreases LD-associated UBXD8 levels through mechanisms that are not yet known, failing to recruit p97/VCP to the LD, which leads to the accumulation of HSD17β13 on the LD. Increased HSD17β13 inhibits LD breakdown, resulting in the accumulation of LD in the liver, which promotes steatosis of ALD.
