Analysis of the effects of a shift from rich to poor carbon during metaphase. Cells were grown overnight in YPD medium and arrested in G1 phase with alpha factor. The cells were then released from the arrest into fresh YPD at 25°C. At the point indicated by the arrow, half of the culture was shifted to poor carbon (bottom panel). Samples were collected at the indicated time points to analyze Clb2 levels by western blot. Samples for mass spectrometry analysis were collected 10 min after the shift to poor carbon.
