Phosphorylation and arrestin binding modulate the production and decay of Meta III in mouse rods. The time course of Meta III was measured as its normalized absorbance (Kolesnikov et al., 2003) after 50% rhodopsin bleach. Fits are bi-exponential functions, f(x) = A(−e-x/τ1 + e-x/τ2), with the following time constants ± SEM. (A) WT: τ1 = 1.5 ± 0.2 min and τ2 = 17 ± 1.2 min (n = 6). (B) Arr1−/−: τ1 = 2.3 ± 0.2 min and τ2 = 27 ± 1.5 min (n = 5). (C) Arr1Ox: τ1 = 2.7 ± 1.1 min and τ2 = 7.6 ± 0.8 min (n = 4). (D) Grk1−/−: τ1 = 0.98 ± 0.1 min and τ2 = 13 ± 0.9 min (n = 6). The error bars represent SEM. All recordings were made at 37°C.
