Schematic summary. (A) Cargo vesicles are delivered to the apical plasma membrane in dependence on Rab11a, Rab8a, and Myo5B. The v-SNARE–like Slp4a is recruited to the vesicle via Rab27, Rab8a, and Rab3b and binds the t-SNARE Stx3 together with the SM protein Munc18-2 to allow the vesicle to fuse with the plasma membrane. The v-SNARE Vamp7 interacts with Rab11a and Stx3. (B) Upon depletion of either Myo5B or Stx3, cargo vesicles are no longer delivered properly.
