G1-induced stimulation of mTOR signaling requires TrkB receptor activation. Hippocampal slices were preincubated with DMSO or ANA-12 (50 µM) for 30 min and then treated with G1 or E2 for an additional 30 min. At the end of treatments, samples were homogenized and processed for Western blotting. Data are presented as the ratio (fold of control) of: (A) PTEN over actin (n = 4–10), (B) phospho-Akt (p-Akt) over total Akt (n = 3–8), (C) phospho-mTOR (p-mTOR) over total mTOR (n = 4–7), and (D) αCaMKII over actin (n = 4–18). *, P < 0.05; **, P < 0.01; ***, P < 0.001, as compared with control; #, P < 0.05; ##, P < 0.01; ###, P < 0.001, as compared with G1 or E2 alone (one- or two-way ANOVA). Some of the actin bands in both A and C look very similar because they show the same data. Images in these two panels come from the same blot, in which different primary and secondary antibodies were used. Thus, the same actin blot serves as loading control for both PTEN and CamKII. They have been separated into two panels for consistency in figure formatting. Black lines indicate that intervening lanes have been spliced out. Error bars indicate means ± SEM.
