Ssd1 localizes to sites of polarized growth. Logarithmically growing cells expressing Ssd1-GFP and Pap1-RFP (a nuclear marker) (FLY3433) were observed by fluorescence microscopy. Ssd1-GFP was enriched in small buds (13–40% of cells, n = 250 cells from four experiments) and bud necks of large budded cells (4–9% of cells, n = 282 cells from four experiments). Ssd1 was not detectable in the nucleus by these methods. Bar, 2 µm.
