A2780 cells were transfected with a non-targeting siRNA (siRNA control [si-con]), an siRNA targeting DGK-α (si-DGK-α) or treated with 10 µM R59022 for 1 h. Cell pellets were collected, and lipids were extracted in the presence of appropriate standards to control for extraction efficiency and to identify the lipids on mass spectrometry. Analysis of each individual molecular species allowed determination of the percentage of each within the total lipid class demonstrating which lipids changed. In each sample, 21 distinct DAG species and 32 distinct PA species were identified and quantified; the species not shown in the table demonstrated minimal changes.
