DCs derived from spinophilin WT and KO mice were loaded with peptide antigen (ovalbumin peptide aa 323–339 or I-Eα 52–68) and then cocultured with CD4+ T cells transgenic for the appropriate TCR (OT-II or DO.11 for ovalbumin or 1H3.1 for I-Eα, respectively). After 18 h, supernatants were isolated and assayed for IL-2 by ELISA. Values shown are the ratio of the response from T cells cocultured with KO/WT DCs. One animal in each group was used for each experiment. The number of experiments (n) is indicated.
