Naive and memory B cells were sort-purified from normal healthy donors (n = 17), STAT3_MUT patients (n = 8), or STAT1_MUT patients (n = 6) and then cultured with CD40L alone or together with IL-21. The levels of secreted IgM, IgG, and IgA were determined by ELISA after 10–12 d. The values represent the mean ± SEM from the indicated number of donors/patients and correspond to the data depicted in Fig. 3 (A and B). Statistical analyses were performed using one-way ANOVA; differences are indicated for normal donors compared with STAT3_MUT or STAT1_MUT B cells.
